Når Syd the Shiba Inu vender tilbake til Shiba Island etter et ti år langt fravær, setter hun i gang en hendelseskjede som velter livet til alle hundene hun hadde etterlatt seg. Hun kommer i kontakt med sin gamle venn, Max igjen og møter en mystisk Labrador-retriever underveis ved navn Quei-Li. (Hva er det et laboratorium gjør på øya?) De er hennes eneste gjenværende allierte når hun prøver å overleve fortiden som fikk henne til å forlate øya i utgangspunktet.
